Hi,
Check the Windows event log for any errors relating to the lockup. Look for ASP.NET warnings or MailEnable errors. The problem does sound like permissions. Perhaps run Process monitor and filter on the w3wp.exe process and then emulate the lockup and inspect the process monitor output for "Access Denied" errors.

We are running four ME servers (different domains) and we have this issue happening on all four of them after upgrading to 9.76 from 9.75 over the weekend.
EDIT: We just rolled one of the servers back to 975 and it resolved the issue. Ian, this has to be something in the 9.76 build. We are on Enterprise Premium BTW.
